Genetic relatedness of Hydrangea macrophylla clones using ISSR markers

摘要: Hydrangea macrophylla is a popular floricultural and garden plant and many new cultivars have been introduced to the market each year. To better figure out their genetic relatedness and potential molecular implication to their morphological characteristics, we selected 10 ISSR primers from our 96-screened primers to investigate the relationships of molecular markers and some morphological traits for nine close related clones of Hydrangea macrophylla. The 207 bands were produced from these 10 primers and average bands per primer were about 21. Among them, 171 bands were polymorphic bands, which accounted for 82.61% of total bands. Hydrangea macrophylla 'Green Mentle' was a bud mutation from H. macrophylla 'Goliath' with greenish tepals, which could be distinguished by 23 unique bands from ' Goliath' using ISSR markers. 'Twist N Shout' is one of the most representative lace cap cultivars, but one of its bud mutations had missed the typical characteristic of lace cap. However, ISSR markers revealed that these two clones just had three different bands from total 207 bands. Double flowers (multiple layers of tepals) were significant different from ISSR markers when compared to single flowers. From 207 bands amplified for both single flowered lace cap cultivar 'Twist N Shout' and double flowered lace cap cultivar 'Izu No Hana', there were only 75 shared bands. ISSR markers provide useful molecular information among the cultivars of bigleaf hydrangea, which can potentially lead to the link between morphological characteristics to their gene(s).
