Metabolic resistance to bensulfuron-methyl endowed by the CdCYP71FL1 gene in Cyperus difformis from China

摘要: Cyperus difformis is a problematic weed in direct-seeded rice fields in China. Due to long-term selection pressure from herbicides, C. difformis has evolved resistance to bensulfuron-methyl (BSM). Our previous study revealed the existence of both target-site gene mutations and metabolic resistance mechanisms in the HN-8 population. However, the specific mechanisms of metabolic resistance were not fully understood and therefore were investigated further. The resistant HN-8 population, compared to the susceptible population, exhibited over-expression of six cytochrome P450 monooxygenases (CYPs), five ATP-binding cassette transporters (ABCs), one UDPglucuronosyltransferases (UGT), and one Glutathione S-transferase (GST). Notably, three CYPs were identified as CdCYP71FL1, CdCYP74B57, and CdCYP94C180, with CdCYP71FL1 being induced by BSM in the HN-8 population. Heterologous expression of CdCYP71FL1 in Arabidopsis thaliana conferred resistance to BSM, bispyribac-sodium, and bentazone, while its expression in yeast facilitated the degradation of BSM. These results indicate that CdCYP71FL1 plays an important role in the metabolic resistance observed in the HN-8 population. Given the complexity of metabolic resistance compared to target-site resistance, it is crucial to prioritize tailored management strategies for herbicide-resistant weeds driven by metabolic mechanisms.
