Comparative genome and transcriptome analysis reveals pathogenic mechanisms of Neoscytalidium dimidiatum in pitaya

摘要: Pitaya canker, caused by Neoscytalidium dimidiatum poses a severe threat to pitaya production; however, its molecular pathogenicity remains poorly understood. In this study, we used whole-genome sequencing, comparative genomics, and transcriptome profiling to elucidate the virulence mechanism of a highly aggressive strain (BH5). The approximate 44 Mb genome revealed a rich repertoire of genes encoding carbohydrate-active enzymes (CAZymes), particularly pectinases, and numerous secondary metabolite genes implicated in toxin synthesis. Comparative analyses with related Botryosphaeriaceae fungi identified unique expansions and contractions in gene families associated with host cell wall degradation and immune evasion. Transcriptome analysis at 12, 24, and 48 h post-inoculation (hpi) revealed marked upregulation of key virulence factors, notably a suite of CAZyme genes involved in polysaccharide degradation, including pectinases, cellulases, and hemicellulases. This expression pattern suggests a coordinated strategy by N. dimidiatum BH5 to break down the complex cell wall polysaccharides of pitaya, thereby facilitating host tissue penetration and colonization while simultaneously undermining plant defense. These results provide new insights into the multifaceted infection strategy of N. dimidiatum BH5 and lay the groundwork for the development of targeted disease management approaches.
